When we integrate site to cloudflare to use it as CDN , is it possible to set that only one edge location is used to serve requests and not others?

No, there would be very little benefit in that over just pointing directly at your origin server.
Enterprise Plans can use the Data Localization feature to control where data is/is not decrypted…
