Adding WPMU subsites

I’ve read the bit about WPMU, but I don’t really understand. I know to add the root domain, but then for the sub-domains, do I need to change the nameservers also? My sub-domains are mapped to their own domain names.

If it is something about changing or adding to the CNAME and A records, can you give me a step-by-step on what to do? I am familiar with changing name servers, but not with CNAME and A records.
